vascular injury, disrupting the blood-brain barrier (BBB) and brain homeostasis. Our study aims to elucidate a role within the brain for TREM-like Transcript-1 (TLT-1), an abundant platelet immunoreceptor that plays <\/span><span class=\"NormalTextRun SCXW54417420 BCX0\">an important role<\/span><span class=\"NormalTextRun SCXW54417420 BCX0\"> in immuno- hemostasis. Following a pro-inflammatory insult such as sepsis, TLT-1 in platelets is cleaved, releasing soluble TLT- 1 (sTLT-1) into the bloodstream as an important biomarker of coagulation. In microvascular brain endothelial cells, sTLT-1 was shown to affect their actin cytoskeleton by increasing cell area. TLT-1\u2019s expression was\u00a0<\/span><span class=\"NormalTextRun SCXW54417420 BCX0\">identified<\/span><span class=\"NormalTextRun SCXW54417420 BCX0\">\u00a0in our lab in cultured brain cells, including astrocytes, microglia, and neurons by western blot, suggesting a\u00a0<\/span><span class=\"NormalTextRun SCXW54417420 BCX0\">possible role<\/span><span class=\"NormalTextRun SCXW54417420 BCX0\">\u00a0for TLT-1 in the central nervous system (CNS). Our studies\u00a0<\/span><span class=\"NormalTextRun SCXW54417420 BCX0\">identified<\/span><span class=\"NormalTextRun SCXW54417420 BCX0\">\u00a0TLT-1 protein expression in rat brain by immunofluorescence, colocalizing with brain cells such as astrocytes. Ongoing experiments are directed towards developing an in vitro BBB model to test the effect of sTLT-1 on BBB permeability using resistance measurements. These studies will examine a physiological function of this immunoreceptor in the CNS and explore its relevance in physiological and pathological states. Research into new molecules involved in neurovascular injury can uncover new avenues for therapeutic intervention, such as strategies for repairing the blood-brain barrier or modulating investigate the importance of Kv1.3 in astrocytes under resting and inflammatory contexts using in vitro models. <\/span><\/p><p>We will determine Kv1.3\u2019s protein expression in wild-type (WT) and Kv1.3 knockout (KO) primary mouse \u00a0astrocytes we will perform Western Blot and Immunohistochemistry (IHC). We will also evaluate how \u00a0Interferon-<em>y<\/em> (IFN-<em>y<\/em>), and PAP-1 changes pSTAT1\/3 levels in primary astrocytes. These experiments will \u00a0allow us to determine astrocytic immune responses involving the Kv1.3 channel after genetic and pharmacological modifications.<\/p><p>Aim 2. <span style=\"text-decoration: underline;\">To analyze an existing proteomic dataset of WT vs Kv1.3 KO mice model.<\/span><\/p><p>We will analyze proteomic data of astrocytes of a neuroinflammatory LPS model in WT vs Kv1.3 KO mice.
